In the post-menopausal women with hormone receptor-positive advanced breast cancer, AASR have been reported in 0.3% of patients treated with placebo and 0.3% of patients treated with tamoxifen. In the intent-to-treat population, nolvadex (Nolvadex) has been shown to have an increased risk of AASR in patients with prior AASR (1-2%), and to increase the risk of AASR in patients with prior AASR. In the adjuvant therapy with letrozole in advanced breast cancer, the relative risk of AASR increased significantly with higher doses (p=0.01). In patients with prior AASR, nolvadex has also been shown to have a greater relative risk of AASR compared with letrozole. In patients with prior AASR, nolvadex is associated with increased side effects (e.g., hot flashes, nausea, dry skin, headache, abdominal pain) and increased mortality. In post-menopausal women with prior AASR, nolvadex has also been shown to have a greater relative risk of AASR compared with letrozole (1-2%), with the increased risk of AASR seen in post-menopausal women with prior AASR.
